From SnOwy - Ed's Wiki Notebook
DNA Clippy
- two homologous regions
- wanted to put two genes, A, B between them
- put the whole thing into a vector
- take construct, put into pk19
- put in insert 1
- put in insert 2
- requirements: insert 1, insert 2 RE sites must not be in any of A, B, or pk19
- which sites should be used to flank the homologous regions?
- some of the pieces of DNA have something selectable
- the iGem workflow takes care of the RE site issue
- request for comments (RFC) -- some are supplementary, some are complementary -- (like a Python PEP)
Mining
- headed toward mining -- is a logical prerequisite of Brandon's idea
- see http://2010.igem.org/Team:Paris_Liliane_Bettencourt/Project/SIP/Downloads -- Paris' database
- intermediate goal is to have a reversed-list for Andre's idea
- Andre's idea: each part is tagged with its previous successful uses
- somebody has to hunt down the curated database discussed at the opening ceremonies (Randy Rettberg)
- alternatively confirm we have to do it ourselves
Duties
- James will look for the mythical curated database that Randy mentioned
- George will look at Paris' SIPs and figure out which are good ontological terms
- DNA Clippy shelved
- SynBioDB shelved until Ed has more time
